Ph.D. Thesis: “Glycan Targeted Gene Delivery to the Dendritic Cell SIGN Receptor”
My research project centered around the development of a novel targeting ligand for non-viral gene delivery. The ligand I prepared contains a high-mannose N-glycan obtained through affinity purification and enzymatic digestion of a natural glycoprotein. The project required the synthesis of an unnatural amino acid used in solid phase peptide synthesis. The N-glycan was conjugated to a unique synthetic peptide, and the resulting glycopeptide vehicle was used to establish receptor mediated gene delivery in cell culture. I showed that the glycopeptide vehicle produced selective gene transfer by targeting the DC-SIGN receptor.
